Evidence
Among the better-supported traditional indications in this collection. Multiple randomised trials and meta-analyses find saffron superior to placebo in mild to moderate depression, and comparable to standard antidepressants in head-to-head trials. Studies are small and heavily concentrated in Iran, which reviewers consistently flag.
Safety
Culinary quantities are benign. High doses cause vomiting, bleeding and, at very high doses, are toxic; historical accounts of poisoning are consistent. Uterine-stimulant effects mean it is avoided in pregnancy at medicinal doses. Adulteration is rampant — it is one of the most faked foods in the world.

Historical use
Depicted in Minoan frescoes at Akrotiri and Knossos around 1600 BCE, including saffron-gathering scenes. Used as spice, dye, perfume and medicine across the ancient Mediterranean and Persia, and consistently associated with mood.
Recorded as history and attributed where possible. Long use establishes that a plant was widely used, not that it worked, and not that it was safe.
Worth knowing
Saffron has the most interesting evidence profile in this collection: a traditional association with mood going back to the Minoans, and a modern trial literature that supports it.
The consistent finding across meta-analyses is superiority to placebo in mild to moderate depression and rough equivalence to SSRIs in direct comparisons. The consistent criticism is that almost every trial comes from Iran, the world's dominant producer, and the effect has not been replicated at scale elsewhere. Both facts are load-bearing.
It is also the most adulterated spice in the world, and the fakes are old — safflower, marigold, dyed maize silk, and simple padding with the yellow styles that carry none of the flavour. Medieval Nuremberg burned people for saffron adulteration under the Safranschou code. A trial result on authenticated Iranian stigmas says very little about a jar bought cheaply.
